Melanotan 2 (MT-2) is a synthetic analogue of alpha-melanocyte-stimulating hormone (α-MSH), a naturally occurring peptide derived from pro-opiomelanocortin (POMC). It is a cyclic heptapeptide that acts as a non-selective agonist at melanocortin receptors (MC1R, MC3R, MC4R, MC5R), studied for its effects on melanogenesis, sexual function, and appetite regulation.
What Is Melanotan 2?
Melanotan 2 was first developed at the University of Arizona. Its predecessor Melanotan I (afamelanotide) has been approved in some jurisdictions as a therapeutic agent for erythropoietic protoporphyria (Scenesse®). Melanotan 2 differs in its broader receptor affinity profile and activity at MC4R — the basis for its study in research areas beyond melanogenesis.
Melanotan 2 Research Background
Melanogenesis research: MT-2’s primary studied mechanism involves activation of MC1R on melanocytes, stimulating melanin production. Preclinical studies in rodent models have documented increased skin pigmentation following administration.
Sexual function research: MT-2’s activity at MC4R receptors in the hypothalamus has made it a subject of sexual function research. The compound PT-141 (bremelanotide) was developed directly from MT-2 research and has since received FDA approval for HSDD in women.
Appetite and metabolic research: MC4R agonism has been associated with appetite suppression in animal models — an active area of investigation in obesity pharmacology.
Photoprotection research: MT-2 has been studied in models of UV-induced skin damage as a potential photoprotective agent.
