Selank is a synthetic heptapeptide anxiolytic and nootropic peptide developed by the Institute of Molecular Genetics of the Russian Academy of Sciences, derived from the endogenous tetrapeptide tuftsin (Thr-Lys-Pro-Arg) with an additional tripeptide sequence conferring stability. It has been investigated in Russian clinical research for anxiety and cognitive function, and holds official drug status in Russia where it is approved as an anxiolytic — making it one of the few research peptides with a formal regulatory approval pathway in any jurisdiction.

Anxiolytic mechanism: Selank’s primary studied mechanism involves modulation of GABA-A receptor expression and enhancement of BDNF (Brain-Derived Neurotrophic Factor) production. Unlike benzodiazepines which act as direct GABA-A positive allosteric modulators, Selank’s mechanism appears regulatory — upregulating endogenous GABAergic tone rather than directly binding the receptor. Russian clinical studies report anxiolytic effects without the sedation, dependence, or cognitive impairment associated with classical GABA-acting anxiolytics.

Cognitive and nootropic research: Studies have examined Selank’s effects on working memory, attention, and learning in animal models and small human trials. Proposed mechanisms include BDNF upregulation, enkephalin metabolism modulation, and serotonergic/dopaminergic system interactions.
